摘要 目的:建立丹芎方(丹参,川芎)中多种有效成分同时定量的RP-HPLC方法。方法:应用高效液相色谱法测定丹芎方中有效成分丹参素、阿魏酸、隐丹参酮和丹参酮ⅡA的浓度。采用Agilent ZORBAX SB-C18(5μm,150mm×4.6mm)色谱柱,以甲醇-0.5%(v/v)冰醋酸溶液为流动相进行梯度洗脱,流速为1mL/min,柱温为室温。丹参素、阿魏酸、隐丹参酮和丹参酮ⅡA的检测波长分别为281,320,254和254nm。结果:丹参素、阿魏酸、隐丹参酮和丹参酮ⅡA在线性范围内回归方程的相关系数均大于0.99992,线性关系良好,4种组分检测精密度RSD〈5%。加样回收率为98.1%~104.05%。结论:通过对不同批次及同一批次不同时间的样品的含量进行测定,说明本方法切实可行,稳定可靠,且重现性好,可作为丹芎制剂的质量控制标准。 AIM: To develop a new method for simultaneously determining four effective components in Danxiong Preseription (Radix et Rhizoma Salviae Miltiorrhizae, Rhizoma Chuanxiong ). METHODS: A RP-HPLC method was set up to determine the concentration of danshensu,ferulie acid, cryptotanshinone and tanshinone ⅡA in Danxiong Prescription. They were successfully separated on an Agilent ZORBAX SB-C18 column(5 μm,150 mm × 4.6 mm) at ambient temperature. A mixture of methanol and 0.5% (v/v) acetic acid in water were used as mobile phase by employing gradient elution at the flow rate of 1.0 mL/min. Danshensu and ferulic acid were detected at 281 and 320 nm,respectively. Cryptotanshinone and tanshinone ⅡA were detected at 254 nm. RESULTS: All the compounds showed good linearity(r 〉0. 999 92) between the peak areas of the componets and their concentration in a relatively wide concentration range. The precision of four components were both below 5%. The recoveries were between 98.1% and 104.05%. CONCLUSION : The method is simple, sensitive, specificity and good reproducibility. It has been successfully applied to quality evaluation of Danxiong Prescription. It can also be used as a quality control method for Danxiong preparation.
