摘要
目的研究 Survivin 基因在原发性肝细胞癌中的表达水平及其与肝癌侵袭转移的相关性。方法收集4种肝细胞癌细胞株,35例原发性肝癌组织标本及相应的癌旁组织,以逆转录-聚合酶链反应(RT-PCR)检测 Survivin mRNA 转录情况;Western blot 法检测 Survivin 蛋白表达。结果在 mRNA 水平4株肝癌细胞株随侵袭转移能力升高 Survivin 表达依次升高(P<0.01),HC-CLM6细胞株中 Survivin 表达水平最高。肝癌组织 Survivin mRNA 表达均为阳性,而癌旁组织内无一例阳性表达,Survivin mRNA 的表达水平在肝内转移组(1.082±0.213)显著高于肝内无转移组(0.799±0.324,P<0.05)。Western blot 的结果与 RT-PCR 结果一致,肝内转移组 Survivin 蛋白表达(0.892±0.198)显著高于肝内无转移组(0.342±0.237,P<0.05)。结论 Survivin 在原发性肝细胞癌中表达,且与侵袭转移等肿瘤的恶性生物学行为有关,Survivin 基因高表达可作为判断 HCC 侵袭性和进展的一个潜在重要指标。
Objective To investigate the expression of Survivin in hepatocellular carcinoma (HCC) and its relationship with the invasion and metastasis of HCC. Methods Four HCC cell lines and 35 HCC specimens with their neighboring noncancerous tissues were obtained. Semi-quantitative RT-PCR was used to detect Survivin mRNA levels. Western blot was used to assay the protein expression of Survivin. Results Survivin mRNA expression was gradually increased with the increase of invasion and metastasis ability in four HCC cell lines ( P 〈 0.01 ). The expression of Survivin mRNA was highest in the cell line HCCLM6. Survivin mRNA expression was detected in HCC tissues. In contrast, no expression of Survivin was detected in corresponding noncancerous tissues. Survivin expression relative amount in cases of intrahepatic metastasis was 1. 082 ± 0.213, higher than that without intrahepatic metastasis ( 0. 799 ± 0. 324, P 〈 0.05 ). The levels of Survivin protein in cases of intrahepatic metastasis (0.892 ± 0. 198 ) were significantly higher than those without intrahepatic metastasis ( 0. 342 ± 0.237, P 〈 0.05 ). Conclusion The expression of Survivin in HCC might have a close relationship with invasion and metastasis. The high expression of Survivin may act as an important index to indicate the process of the invasion and metastasis in HCC.
