摘要
运用RAPD技术对核桃与铁核桃特征性条带进行标记。分别利用8个核桃样品和8个铁核桃样品构建2个物种的基因池,用327个随机引物,选出10个多态性引物,进一步用这10个引物进行单株确认,发现只有引物D6在核桃类群的8个样品中扩增出一条250bp的特异带,而在铁核桃类群的8个样品中无此特异带。初步分析认为,D6-250可能是核桃与铁核桃之间的特异性分子标记,该结果可为核桃和铁核桃种质鉴定提供一定的理论依据。
Random amplified polymorphic DNA (RAPD) was applied to detect the relationship at the species level between Juglans regia L. and J . sigiUata Dode. Both J . regia DNA- pool and J. sigillata DNA- pool were set up by using 8 J. regia samples and 8 J. sigillata samples, respectively. 327 primers were screened, of which 10 polymorphic primers were identified. Two DNA- pools and all individuals were verified by using the 10 polymorphic primers three times. A 250 bp DNA band was amplified only from primer in D6 J. regia DNA- pool and 8 corresponding samples of J. regia, but absent in both J. sigillata DNA - pool and 8 corresponding samples of J. sigillata. The results tentatively showed that D6-250 was a special molecular marker related to J. regia and J. sigiUata at the species level. This marker can provide rome basis for the relationship identification of germplasm resources of the two species.
