摘要
以八眉猪为试验材料,提取其内脏脂肪、皮下脂肪、肌肉、肾脏、肝脏、心脏、肺脏和脾脏总RNA和总蛋白质,设计合成PPAR-γ1和PPAR-γ2基因的引物,以猪β-actin基因为内参,用半定量(Semi-Quantitative,SQ)RT-PCR检测PPAR-γ1和PPAR-γ2 mRNA在以上各组织中的表达;用免疫印迹技术(Western Blot)检测以上各组织中PPAR-γ1和PPAR-γ2蛋白的表达。结果显示,PPAR-γ1 mRNA及蛋白在以上各组织均表达,在内脏脂肪和皮下脂肪组织中表达水平显著高于其他组织,在肺脏中表达水平最低;PPAR-γ2 mRNA和蛋白质高表达于内脏脂肪、皮下脂肪、脾脏和心脏,而在肺脏、肾脏、肝脏、肌肉中表达水平很低。结果提示,PPAR-γ在不同组织的表达差异,可能与其在不同组织中的功能有关。
Total RNA and protein were extracted from visceral adipose tissue, subcutaneous adipose tissue, skeletal muscle, kidney, liver, heart, lung and spleen of Bamei pig. The primers of PPAR-γ1 and PPAR-γ2 were designed and synthesized. The specific expression of PPAR-γ1 and PPAR-γ2 in different tissues were investigated by Semi-Quantitative RT-PCR and Western Blot. The results showed that the mRNA and protein of PPAR-γ1 were expressed in all above-mentioned tissues, and highly expressed in visceral adipose tissue, subcutaneous adipose tissue and lower expressed in lung; PPAR-γ2 mRNA and protein were highly expressed in visceral adipose tissue, subcutaneous adipose tissue, spleen and heart, while remarkable lower in lung, kidney, liver and muscle. In conclusion, the results suggested that PPAR-γ might have various effects on different tissues of pig.
