Survivin在子宫内膜癌中的表达及其与细胞凋亡的相关性研究

Expression of Survivin protein in the endometrial carcinoma and its relation with apoptosis

目的探讨凋亡调控基因Survivin在子宫内膜腺癌发生发展中的作用及其与细胞凋亡的相关性。方法应用免疫组化方法检测Survivin蛋白在正常、增生性子宫内膜和子宫内膜癌中的表达,应用Tunel法检测凋亡细胞。结果(1)在正常增殖期宫内膜中,腺上皮细胞的凋亡较为罕见,从简单型增生到子宫内膜癌凋亡指数呈上升趋势。在子宫内膜癌中,凋亡指数AI与组织病理学分级呈正相关。(2)从正常子宫内膜到子宫内膜癌,Survivin蛋白表达的阳性率逐渐增加。在子宫内膜癌中,随着组织学分级、手术分期的升高,survivin的阳性率、过表达率增加,并有显著性差异。(3)Survivin的表达与凋亡指数间无显著相关性(r=0.200,P≥0.05)。结论Survivin可能通过抑制凋亡并促进细胞增殖参与了子宫内膜癌发生发展。细胞凋亡在子宫内膜癌中的增高可能反映了肿瘤的恶性程度。

Objective to investigate the expression of Survivin in normal, proliferative and endometrial carcinoma and its relationship with apoptosis. Methods Immunohistochemical SP method was performed to detect the expression of Survivin protein. The apoptotic glandular cells were examined by TUNEL method. Results （1） The incidence of apoptosis was low in normal endometrium, but it increases from simple endometrial hyperplasia to complex hyperplasia and atypical endometrial hyperplasia, and markedly increases in endometrial carcinoma. In endometrial carcinoma, the increase of Al was correlated with the histological differentiation （P〈0.05）. （2）The rate of Survivin increased progressively from normal endometrium and atypical hyperplasia to carcinoma. With endometfial adenocarcinomatous histological grade and surgical-pathological stage increasing, Survivin expression-rate and overexpression-rate increased, and the differences were significant （P〈0.05）. （3）Survivin did not show any statistical significant correlation with AI. Conclusions Survivin may involved in the progression of endometrial carcinoma as a result of the inhibition of cell death and promotion of proliferation, and the increase of AI in the endometfial carcinoma may reflect the extent of malignance in tumor.
