摘要
为了探讨脑缺血后血小板活化因子(platelet-activatingfactor,PAF)含量的变化,采用沙土鼠单侧脑缺血和大鼠局部脑缺血模型,用薄层层析(TLC)结合生物法测定了缺血后脑组织PAF的含量结果:(1)沙土鼠右侧颈总动脉结扎3h再灌2h,右侧大脑组织PAF含量较左侧明显升高,分别为(440±090)和(130±042)pg/g(P<005);(2)大鼠左侧大脑中动脉(MCA)凝闭后48h,左侧大脑组织PAF含量右侧亦明显升高。
To explore the release of platelet-activating factors (PAF), an ischemia model was established with the unilateral stroke in gerbils and with the regional cerebral ischemia in rats. Thin layer chromatography (TLC) was combined with biological methods for the determination of PAF in the cerebral tissue after the ischemia. Result: 1) cerebral tissue PAF content was significantly higher in the right hemisphere than in the left hemisphere, respectively(4.40+0.90)and(1.30+0.40)pg/g (P<0.05), after 2-hour reperfusion preceded by 3-hour ligation at the right carotic arteria of gerbils; 2) cerebral tissue PAF content was significantly higher in the right hemisphere than in the left hemisphere, respectively(5.74+0.74)and(0.76+0.05)pg/g (P<0.05), 48 hours after the occlusion at the left middle cerebral artery (MCA) of rats. In the light of the results a analysis was devoted to the possible PAF role in the etiological mechanism of neuron injury coming after cerebral ischemia.
