Significant organs damage induced by high blood pressure variability in SAD rats

摘要 目的:研究去窦弓神经(SAD)大鼠的血压、血压波动性(BPV)和器官损伤及它们之间的相互关系。方法:应用计算机化清醒自由活动大鼠血流动力学监测技术对32周龄的SAD大鼠进行24h动态血压监测。数据经计算机实时和脱机分析后得到24h血压、收缩压波动性(SBPV)、舒张压波动性(DBPV)等参数。将动物处死,取心脏、肾脏、大脑等器官病理切片,分别行一般和特殊染色。参照预先制定的评分标准,根据所观察器官大体和镜下变化,给每一只动物以相应的评分作为器官损伤程度的半量化指标。将血压及BPV与器官损伤评分做相关性分析。结果和结论:(1)SAD大鼠的24h长、短期及总SBPV和DBPV明显高于假手术对照组;(2)SAD大鼠的器官损伤明显重于假手术对照组,并且这种损伤与其长期BPV和总BPV的提高有正相关性。 Objective: To study the relationship between blood pressure (BP), blood pressure variability (BPV) and organs damage in SAD rats at 32 weeks of age. Methods: Intra arterial blood pressure was continuously recorded for 24 h in freely moving SAD rats by micro computer system. On line and off line computer analysis of the BP curve allowed calculation of BP and BPV. Hearts, kidneys,brains and spleens were excised, fixed, dehydrated and embeded in paraffin. Sections were stained with HE and special methods separately to assess the severity of organs damage. Organs damage was semiquantified according to predetermined criteria. Results and Conclusion:(1)24 h long term , short term and total SBPV and DBPV were higher in SAD rats than those in control rats [SBPV: (1.2±0.5),(1.5±0.5), (1.9±0.5) vs (0.7±0.2), (1.2±0.2), (1.4±0.2) kPa,P<0.01;DBPV: (0.9±0.3), (1.5±0.3), (1.8±0.3) vs (0.7±0.5), (1.3±0.2), (1.6±0.2)kPa, P<0.01)]. (2) Organs damage scores were higher in SAD rats than those in control rats [(7.6±2.44 )vs (3.6±1.59),P<0.001] and organs damage was positively correlated with 24 h long term and total BPV in SAD rats.
