摘要
目的为探讨γ射线对犬bcl-2基因的影响及意义。方法将^103钯(^103Pd)放射性支架和普通支架分别植入犬的肝外胆管,免疫组化方法检测γ射线诱导的犬胆管壁凋亡增殖平滑肌细胞中的bcl-2表达,计数凋亡细胞,计算机图像分析系统检测两组胆管管腔面积。结果bcl-2表达^103Pd支架组较普通支架组为弱,胆管出现增殖平滑肌细胞明显凋亡;肝外胆管无明显狭窄;普通支架组未出现增殖平滑肌细胞明显凋亡,肝外胆管有明显狭窄。结论bcl-2表达水平与细胞凋亡发生和细胞对辐射敏感性有关;^103Pd放射性支架通过降低bcl-2表达,促进胆管增殖平滑肌细胞凋亡,从而抑制肝外胆管狭窄。
Objective To study the expression and significance of bcl-2 gene in the apoptotic muscle cells in γ-radiation-induced muscle cell lines. Methods bcl-2 gene in the control and r-radiation-induced apoptotic muscle cells was analyzed by using immunohistochemical technique. Results The bcl-2 gene transcript more highly expressed in general stent dog bile duct than in ^103Pd radioactive stent dog bile duct, but apoptotic muscle cells were more highly in ^103 Pd radioactive stent dog bile duct than in general stent dog bile duct. Conclusion The low level of expression of bcl-2 gene may aid the effects to understand the muscle cell sensitivity to γ-radiation apoptosis. ^103Pd radioactive stent may reduce the expression of bcl-2 gene in dog bile duct and prevent the biliary narrowing after dog bile duct injury by balloon.
