Screening for Antifoam with High Performances Based on Its Physical and Physiological Effects on the Fermentation of Biotechmycin

摘要 综合消泡剂的物理消泡效果及其对发酵过程参数的影响2个主要方面,考察4类(共13种)消泡剂对必特螺旋霉素发酵的影响。首先从4类消泡剂中筛选出7种物理效果好的消泡剂,再通过考察这7种消泡剂对必特螺旋霉素生物合成的影响,最后筛选到了消泡、抑泡效果好,并对发酵有显著促进作用的#12消泡剂。在此基础上研究这种消泡剂对发酵过程参数的影响,并优化它的添加浓度,最终使发酵单位提高34.5%。 Regarding to antifoam used in fermentation process to control foam level,there have been many investigation reports up to now,which mainly focus on their physical effects such as the defoaming and foam suppression,however,there have been few literatures on effects of antifoam on physiological state of antibiotic producer,especially on the antibiotic biosynthesis.In the present study,13 antifoams covering 4 types were investigated base on theirs physical and physiological effects on the biotechmycin production.Firstly,seven antifoams among 13 antifoams tested were selected based on their high physical effects on foam formation of the fermentation broth.The optimum antifoam #12 was then obtained based on not only its high antifoam capability,but also its improvement of biotechmycin biosynthesis by Streptomyces spiramyceticus WSJ-1-195.Furthermore,#12 antifoam concentration was optimized in the medium formula,and the titer of biotechmycin was increased by 34.5% compared with the control.
