摘要
以农杆菌介导法转几丁质酶基因和蝎毒蛋白基因的晋4和H165油菜种子为试材。采用PCR检测、农艺性状调查、田间抗虫鉴定等技术,对转基因植株及其后代进行了选育研究。T1、T2和T3代材料的PCR检测结果显示目的基因在转基因植株及其后代中可稳定遗传;抗虫鉴定结果证明转基因油菜植株的抗虫性得到提高,转基因株系的被害率、被害程度明显小于相应对照,转基因株系中菜粉蝶幼虫的存活率低于对照,而且存活虫的发育进程比对照缓慢;农艺性状调查结果证实,转基因手段对油菜植株的角果数、角粒数和千粒重等性状没有太大影响。根据上述结果,在T3代时,筛选得到J5-1J、5-2、H5-1和H11-1四个纯合的转基因油菜株系。
In this study,the transgenic seeds used as material were obtained from insect-resistant transgenic oilseed lines that transformed by Agrobacteria-mediated approach on Jin 4 and H165 (Brassica napus).The results of PCR amplification for T1,T2 and T3 progenies showed that chitinase gene and scorpion toxin gene can stablely inherited. Biological assay for insect-resistant confirmed that the insect-resistance of transgenic plants increased,The percentage of injury and the level of damage of transgenic plants were much less than control plants. Compared with control plants,the survival number of Diamonback moth larvas in transgenic pa lnts were less ,too. Furthermore,the development of survival insects were slowly. Investigation results of transgenic plants on agronomic characters (pod number/plant,kernel number/pod,thousand kernels weight)indicated:no considerable difference existed between transgenic and control plants. Based on above results,4 transgenic pure lines,J5-2,J5-2,H5-1,H11-1,were obtained from T3 generation transgenic line by selection breeding.
