摘要
以优良光温敏核不育系Y58S及其杂交组合Y两优1号和生产中广泛应用的部分光温敏核不育系和恢复系等18个水稻品种为材料,利用SSR分子标记进行了DNA指纹图谱的构建。聚类分析表明,18个供试水稻品种的遗传相似度为0.672-0.945;利用筛选到的RM164、RM18和RM258等3对引物扩增出的14条多态性片段初步构建了18个供试水稻品种的DNA指纹图谱,每个品种的DNA指纹图谱具有特异性,可互相鉴别;引物RM164可在超级杂交稻组合Y两优1号、两优培九中扩增出父母本的互补带型,从而可鉴别其杂交种子中混杂的母本杂株。
The DNA fingerprinting of 18 two-line hybrid rice varieties and their related parents, including Y58S and its hybrid Y Liangyou 1 and some other important PTGMS lines and restorer lines widely used in commercial production, was conducted with 60 SSR markers. The cluster analysis results showed that the genetic similarity of all tested hybrid rice varieties/parents ranged from 0. 672 - 0. 945 based on the data of 12 SSR primers selected from the tested 60 primers. The DNA fingerprinting of all of the tested varieties/parents was primarily established with three primers including RM164, RM18 and RM 258, by which 14 DNA bands were stably and repeatedly amplified among 18 tested varieties/lines and each variety/parent had its unique DNA fingerprinting pattern so that they could be distinguished from each other. Moreover, the primer RM164 made a perfect male and female parental complementary amplification of two super hybrid rice varieties Y Liangyou 1 and Liangyou Peijiu, indicating that it can be used to distinguish female seeds from F1 hybrid seeds of these two hybrids.
