摘要
目的建立紫草软胶囊中左旋紫草素的含量测定方法。方法采用HPLC法,考证于不同流动相组合、流速及柱温等条件下测定紫草软胶囊中左旋紫草素的效果。结果以DIKMAODS C18(250mm×4.6mm,5μm)色谱柱,以甲醇-水-甲酸(85:15:0.5)为流动相,流速1.0ml·min^-1,波长516nm,柱温25℃,进样量100μl。检测紫草软胶囊中左旋紫草素在10.2-35.3μg·ml^-1浓度范围内线形关系良好(r=0.9997);平均回收率为100.6%,RSD=1.83%(n=5)。结论所建立的HPLC实验方法简便、检测结果准确,可用于紫草软胶囊的质量控制和评价。
Objective To establish a method for the determination of content of shikonnin in Radix arnebiae soft capsules by HPLC. Methods The content of shikonin was determined by HPLC, the chromatographic conditions were DIKMAODS C18 column ( 5μm, 250mm×4.6mm), mobile phase methanol - water - Formic Acid ( 85 : 15 : 0.5 ), detection wavelength 516 nm, temperature 25 ℃, flow rate 1.0ml·min^-1 with the load of 100μl. Results It keeps a good linear when the shikonin was 10.2-35.31μg·ml^-1 (r=0.9997), and the average recovery rate was 100.6 % , RSD =1.83%. Conclusion This method was a simple, accurate one with a good reproducibility that can be used for the quality control in assessmen of the quality of Radix amebiae soft capsule.
