银杏达莫对兔心肌缺血再灌注诱导内皮细胞损伤的影响及机制 被引量:11

The Effects and Mechanism of Extract of Ginkgo Biloba on Endothelial Cells Injury Induced by Myocardial Ischemia-reperfusion in Rabbits

摘要 目的研究银杏达莫对兔心肌缺血再灌注损伤内皮细胞的保护作用。方法结扎兔左冠状动脉前降支制作心肌缺血再灌注损伤模型。实验兔随机分为空白对照组(假手术)、缺血再灌注组和银杏达莫组。银杏达莫组在心肌缺血前10min给予银杏达莫。观察各组缺血再灌注过程中心电图的动态改变,以及银杏达莫对血管性血友病因子及丙二醛和血清一氧化氮含量的影响。结果缺血再灌注组和银杏达莫组均造成明显的心电图动态改变,与空白对照组比较,银杏达莫组心电图ST段出现有效改变。银杏达莫组与缺血再灌注组比较血管性血友病因子活性和丙二醛水平较缺血再灌注组显著降低,而血清一氧化氮水平明显提高。结论银杏达莫对心肌缺血再灌注损伤内皮细胞有明显的保护作用,其作用与清除氧自由基的膜脂质过氧化及扩张冠脉有关。 Objective To study the protective effects and mechanism of Egb on endothelial cell injury induced by myecardium experienced isehemia - reperfusion in rabbits. Methods The model of myocardial ischemia - reperfusion injury was established by ligating the left anterior descending artery of New Zealand rabbits. Anesthetized rabbits were randomly divided into 3 groups: blank control group ( n = 9 ) in which left anterior descending coronary, artery was exposed and a piece of silk thread was placed around the artery but not tied ; Myocardial ischemia - reperfusion group ( n = 9 ) in which 1 ml/kg normal saline was injected into vein 10 min before logation. Egb group (n =9) received intravenous inject on of Egb 10min before logation. The effect of egb on the yon willebrand factor (vWF) , malondialdehyde (MDA) in the plasma, the level of serum NO in injured rabbits and ECG were observed. Results Compared with blank control group or myocardial ischemia - reperfusion group, Egb group showed protection against injury as evidenced by more effective improvement in ST of ECG. Egb could evidently decrease the conteets of MDA and vWF and increase serum NO as compared with isehemia - reperfusion group. Conclusion Egb has a protective effect on endordnelial injury induced by myocardial isehemia - reperfusion tbrongh inhibiting lipid peroxidation and increasing NO preduetion.
