摘要
本法系在Cohn和Lyle的荧光分光光度法的基础上,以10%三氯醋酸代替25%HPO_3在常温(18~30℃)下去蛋白,并以pH8.3磷酸盐缓冲液代替pH8.0的该液。本法的重复性试验和回收试验结果满意,最低检出限为25ng/ml,标准曲线至少在2.5μg/ml内呈线性。用本法测定39例健康献血员,血浆GSH值为15.125±3.383μmol/L,男女性别无明显差异。本法灵敏、准确可靠、操作简单、标本用量少。
An improved fluorometric method for measurement of plasma glutathione is introduced. 10% trich loroacetic acid and pH 8.3 phosphate buffer solution were used instead of 25% HPO3 and pH 8.0 phosphate buffer solution respectively and the method might be performed under room temperature. The repetition tests and recovery tests of the method were favorable giving a sensitivity of 25ng /ml and a linear standard curve to at least 2500ng/ml. Plasma glutathione determined by this method in 39 healthy blood donators showed a result of 15.125± 3, 38μmol/L and no any sexual difference. The method is sensitve, accurate and less expensive.
