摘要
为了建立新疆巴旦杏中苦杏仁苷的HPLC定量测定方法,采用乙醇回流提取法提取苦杏仁苷,以ZirchromC18色谱柱(250 mm×4.6 mm,10μm)为固定相,甲醇∶水∶冰醋酸(20∶80∶0.05)为流动相,流速为1.0mL/min,检测波长为210 nm,柱温为35℃。测定结果显示,苦杏仁苷在20.0-70.0μg/mL成良好的线性关系,r=0.999 9,日内、日间RSD均为0.59%(n=5),平均回收率99.6%(n=6)。研究表明,该方法灵敏度高,结果准确,重现性好,方法简便可行,适用于新疆巴旦杏中苦杏仁苷的含量测定,并为其质量控制提供理论依据。
To establish the HPLC method for quantitative analysis of amygdalin in Xinjiang Amygdalus persica L.,Amygdalin was extracted by ethyl alcohol reflux method.The content of amygdalin was determined by use of the HPLC method. In the test,stationary phase was Zirchrom C18 column(250 mm×4.6 mm,10 μm),the mobile phase was a combination of methanol,water,glacial acetic acid in the volume ration of 20∶80∶05,the flow velocity was 1.0 mL/min,the wavelength of UV detector was 225 nm and the column temperature was 35 ℃.The result showed that the linear range of amygdalin was 20.0~70.0 μg/mL,r=0.999 9,RSD both in a few days and in the daytime was 0.59%(n=5),the average recovery rate of sample was 99.6%(n=6).Because of a good saitivity,accuracy,reproducibility and convenient,the method can be used for assaying of amygdalin in Xinjiang Amygdalus persica L.,and provided a theoretical base for its quality control.
