摘要
目的确定水通道蛋白-1、4(AQP1、4)在正常大鼠角膜和晶状体生长发育过程中的表达及分布。方法用免疫组织化学方法观察AQP1、4分别在胚胎13、15、18、20d及出生当天、出生后7d、14d和8周成年大鼠角膜及晶状体中的表达。每组选取3只Wistar大鼠作为实验对象。结果AQP1在胚胎15d大鼠的角膜内皮和晶状体上皮就有表达,随着大鼠的生长发育,表达呈动态变化。AQP4在生后7d大鼠的角膜上皮、晶状体上皮和晶状体纤维中有弱阳性表达,并持续至成年鼠。结论AQP1、4在大鼠角膜和晶状体的表达随着角膜和晶状体的生长发育而调节,此蛋白对维持角膜和晶状体的透明性及正常生理功能发挥了重要作用。
Objective Aquaporin (AQP) is selective water permeability of the inner membrane protein. It is the main internal protein(MIP) members of the family and widely distributed in animals, plants, micro-organisms. This study was to confirm the expression and distribution of AQP1 and AQP4 during the development of cornea and lens of normal rat. Methods Cornea and lens of rat eyes were dissected from rats of embryonic day(E) 13,E15, El8,and E20,postnatal day( P)0, P7,and Pl4,and postnatal week 8, and expressions of AQP1 and AQP4 in cornea and lens were detected using immunohistoehemieal technique with AQP isoform-speeifie antibodies. Results AQP1 was detected in immature corneal endothelium and lens epithelial cells in E15 rat. The expression level of AQP1 in corneal endothelium and lens epithelial cell was gradually increased with the development of rat until E20. Thereafter, AQP1 was expressed in a stable level in cornea and lens of rats. No AQP1 was detected in corneal epithelium of rat. In contrast, AQP4 was firstly found in developing cornea epithelium,lens epithelial cells and lens fiber in P7 rat and presented a faint expression lasting throughout development up to adulthood. Conclusion The expression of AQP1 and AQP4 are regulated along with cornea and lens development. These two proteins are important for keeping physiology balance and transparency of cornea and lens after birth.
