摘要
目的:考察不同产地黄芪中黄芪甲苷的含量。方法:采用高效液相色谱-蒸发光散射法(HPLC—ELSD)测定黄芪甲苷的含量。色谱条件:kromasilC18色谱柱(4.6mm×250mm,5μm);流动相:乙腈-水(38:62);流速:1.0mL·min-1;柱温:室温;进样量:10μL。ELSD检测器条件:漂移管温度:45℃;载气压力:2.02Bar。结果:方法学考察结果表明,黄芪甲苷的线性范围为2.p10.0μg(r=0.9995);精密度实验的RSD为1.30%;24h内稳定性试验的RsD分别为1.71%;重复性试验的RsD分别为1.28%;加样回收率为102.10%,RSD为1.56%。结论:该方法测定结果准确稳定,重现性好;各产地黄芪中黄芪甲苷元含量间变异较大,其中甘肃产黄芪平均质量较优。
Objective: To determine the content of astragaloside IV in Radix astragali from various habit- ats. Methods : The content of astragaloside Ⅳ was determined by high-performance liquid chromatography with evaporative light-scattering detection ( HPLC-ELSD ); The chromatography condition was as follows: Kromasil C 18 column (4.6mm × 250mm, 5μ m) ; mobile phase was CH3 CN-H20(38 : 62); flow speed was 1.0 mL · min-1; temperature of column was room temperature; and the injection volume was 10 μ L.The ELSD conditions were as follows: the temperature of drift tube was 45 ℃, the gas pressure was 2.02 bar. Results : The calibration curveswere linear in the range of 2.0-10.0 μ g(r=0.9995). The precision (RSD) was 1.30%. The stability (RSD) was 1.71% in 24 h and the recurrence(RSD) was 1.28%. The recovery was 102.10% (RSD=1.56%) for astragaloside IV. Conclusion : HPLC-ELSD is a simple and reliable method to determine the content of astragaloside IV in Radix astragali. The content of astragaloside IV in Radix astragali had great changes between the various habitats, the content of astragaloside IV in gansu Radix astragali were the most.
