摘要
目的:建立HPLC法快速测定茶叶中的咖啡因。方法:甲醇+水=70+30(v/v);Agilent-C18柱(4.6 mm×300 mm,5μm);流速1.0 mL/min,柱温30℃,检测波长286 nm,进样量10μL。结果:咖啡因在10~125μg/mL时,浓度与峰面积呈良好的线性关系(r=0.9996),加标回收率为98.40%~101.07%,最低检出限为1μg/mL,相对标准偏差RSD为0.08%~1.32%。结论:用该方法测定茶叶中的咖啡因,操作简单、快速、准确,结果令人满意。
Objective:To establish a rapid method by HPLC in the determination of caffeine concentration of tea.Methods:Chromatographic conditions included Agilent-C18 column(4.6 mm×300 mm,5 μm) and the mobile phase consisting of a mixture of methanol and water by 70+30(v/v).The flow rate was 1.0 mL/min.Column temperature was at 30℃.Detection wavelength was at 286 nm.The sample dose was 10 μL.Results:When the concentration of caffeine was at 10 μg/mL to 125 μg/mL,the perfect linearity between concentration and peak area ratio was obtained(r=0.9996).The recovery rate was 98.40% to 101.07%,and the detection limit was 1 μg/mL,the relative standard deviation(RSD) was 0.08% to 1.32%.Conclusion:This method is simple,rapid and accurate in the determination of caffeine concentrations of tea and satisfactory results could be obtained.
