摘要
目的探讨鱼腥草对甲1型流感病毒核蛋白基因的作用。方法在甲1型流感病毒核蛋白基因保守区设计引物,对其进行筛选,同时对荧光定量逆转录聚合酶链反应(RT-PCR)条件和反应体系进行优化。设病毒对照组、药物对照组、细胞对照组和实验组,采用荧光定量RT-PCR法测定鱼腥草对甲1型流感病毒基因的效应。结果病毒对照组细胞出现细胞病变(CPE),血凝滴度为阳性,其他组无CPE,血凝滴度均为阴性。以荧光定量RT-PCR法重复5次取平均值,实验组RNA测定结果为(2.6±0.8)×106拷贝数/μgRNA,对照组为(2.9±1.2)×109拷贝数/μg RNA,两组比较差异有统计学意义(P<0.01)。结论鱼腥草可抑制甲1型流感病毒核蛋白基因的表达。
Objective To investigate the effect of Yuxingcao (Houttuynia cordata) on influenza A1 virus necleoprotein gene. Methods The specific primers were designed in the conserved region of influenza A1 virus necleoprotein gene, then gene was screened. The reaction conditions and system for fluorescence quantitative polymerase chain reaction ( FQ - PCR) were optimized. The virus control, drug control, cell control and trial groups were designed to detect the effect of Yuxingcao on influenza A1 virus necleoprotein gene by FQ -PCR. Results There occurred cytopathic -effect (CP) and the haemagglutinin (HA) titer was positive in the virus control group, but no CP occurred and HA titer was negative in all the rest groups. The average level of 5 times duplicates by FQ - PCR was 2.93 × 10. 9 copies/μg RNA in the trial group and 2. 60 × 10. 6 copies/μg in the drug control group respectively, the difference between the two groups was significant ( P 〈 0. 01 ). Conclusion Yuxingcao could inhibit the expression of influenza A1 virus necleoprotein gene.
