摘要
目的分析我国特有的三个小型猪品系巴马小型猪、中国农大小型猪、五指山小型猪,过氧化物酶体增殖物激活受体γ协同激活子1基因intron8-exon10序列单核苷酸多态性(SNPs)分布特点,为我国小型猪在糖尿病和代谢性疾病研究中提供基础资料。方法以三个品系小型猪基因组DNA为模板,应用特异性引物进行PCR扩增,扩增产物纯化测序,进行BLAST比对分析。结果在PGC-1基因intron8-exon10这段扩增序列上存在7个单核苷酸多态位点,分别为:1534C→G,1546G→A,1589位插入T,1648C→T,1 653位插入A,1 660位插入G或T,1 830G→A,这些多态位点均位于非编码区,其中,有三个位点发生了单碱基的插入。结论小型猪品种不同多态位点分布有很大差异。
Objective To analyze the single nucleotide polymorphism(SNPs) of peroxisome proliferator activated receptor-γ coactivator 1 gene intron 8-exon 10 in three strains Chinese minipigs,Bama minipigs,Wuzhishan minipigs,and Nongda minipigs,to provide basic data for their use in research of diabetes mellitus and metabolic diseases.Methods Genomic DNA was extracted from blood samples of the minipigs,and served as templates for target gene segment amplified and selected by polymerase chain reaction(PCR).The PCR products were sequenced and analyzed for documenting single-nucleotide polymorphisms(SNPs).Results Seven polymorphisms were found in the target region,1534C→G,1546G→A,T insertion into 1589,1648C→T,A insertion into 1653,G or T insertion into 1660,1830G→A,and all the variations were found in the noncoding region.Conclusion There are variations of SNPs in PGC-1 gene exon 8 existing among the strains and individuals in the three native minipigs.
