经气管/静脉途径行肺脏腺病毒载体基因转染的效率和毒性分析 被引量:3

摘要 目的:分别经气管和静脉途径转染腺病毒载体基因至肺组织,探索不同给药途径对肺组织的转染效率和腺病毒载体转基因治疗的安全性。方法:分别通过气管内滴入和静脉注射途径向家兔转染携带绿色荧光蛋白(GFP)基因的腺病毒载体(Ad-GFP 2×109pfu),于第3 d、7 d、14 d、21 d处死动物,荧光显微镜下观察测肺、肝脏、心脏组织GFP表达,HE染色观察组织病理改变。结果:气管转染组第3 d仅在肺组织内表达GFP,持续21d,且强度大;静脉注射转染组第3 d可在肺、肝脏、心脏组织测及GFP表达,14 d皆消失,强度较弱。HE染色显示,气管转染组第3 d可见肺脏炎性反应,第14 d炎性反应消失,肝脏、心脏均未见炎性反应;静脉注射转染组第3 d可见肺脏、心脏及肝脏内有轻度炎性反应,7 d后炎性反应皆消失。结论:肺脏经气管途径转染腺病毒载体具有较高的靶向性和较长时间基因表达的优点,虽可造成肺部一过性免疫炎性反应,仍不失为肺脏转基因治疗的有效和相对安全的途径。
