仿刺参酸性磷酸酶的提取及粗酶性质研究 被引量:8

摘要 试验结果表明,仿刺参酸性磷酸酶的最佳提取缓冲液为pH 7.5的Tris-HCl缓冲液,最佳沉淀条件为饱和度为80%的硫酸铵。仿刺参体壁和肠道中的酸性磷酸酶最适反应条件均为:40℃,pH4.0。Ca2+、Mg2+和Mn2+对体壁酸性磷酸酶有激活作用,而Fe3+、Fe2+、Zn2+、Cu2+、Ba2+对其有抑制作用,其中Cu2+的抑制作用最强;肠道酸性磷酸酶表现的特征与之类似,但Zn2+的抑制作用最强。PAGE电泳结果表明,仿刺参体壁和肠道中的酸性磷酸酶类型不同,肠道中有3种同工酶。 Extraction and properties of acid phosphatase were investigated from sea cucumber(Apostichopus japonicus). Tris-HCl buffer(pH 7.5)was found to be the optimal extraction solution and 80% of sat- urated (NH4)2SO4 for optimum precipitation. The optimal temperature and pH of acid phosphatase were 40 ℃ ,and 4.0 in body wall and gut. The crude acid phosphatase from body wall was activated by Ca^2+ , Mg^2+ and Mn^2+ , and inhibited by Fe^3+ , Fe^2+ , Zn^2+ , Cu^2+ and Ba^2+ , in which Cu^2+ was the strongest inhibitor. The crude acid phosphatase from the gut had nearly the same property as that from the body walls, and Zn^2+ showed the strongest inhibition. The PAGE revealed that the acid phosphatases in the body walls were different from those in the guts, in which there were three isoenzymes of acid phosphatases.
