摘要 对鱿鱼眼提取透明质酸的初提取液的去蛋白酶解工艺进行了研究。分别对酶种类、酶用量、酶解温度、酶解时间进行单因素实验,得出初步结论后,再采用正交实验对酶解条件进行优化。结果显示,中性蛋白酶去蛋白效果好,且分子量损失最小。酶解温度对去蛋白率影响最大,酶用量对分子量损失率影响最大。鱼眼初提取液的去蛋白酶解最佳条件为:中性蛋白酶用量1.0%、酶解温度50℃、酶解时间6h。该条件下,透明质酸得率85.7%,分子量损失率10.6%,蛋白去除率91.1%。 The enzymatic hydrolysis conditions of hyaluronic acid's primary extract from squid eyes were studied. Firstly, we took single factor analysis of enzyme species, concentration, temperature and reacting time of enzymatic reaction system,to get the elementary conclusions.Then orthogonal experiments were taken to get the optimum medium for enzyme production.The optimal conditions of enzymatic treatment were obtained as follows:neutral protease was selected,enzyme concentration was 1.0%,the reaction temperature was 50℃ and reaction time was 6h. Under these conditions,the yield rat of hyaluronic acid was 85.7% ,molecular weight loss rate was 10.6% ,and protein rejection was 91.1%.
