摘要
以局部血管通透性的改变为指标,比较了旧结核菌素(OT)和牛血清白蛋白(BSA)诱导小鼠迟发型超敏反应(DTH)的作用,并以此为基础,在成模前给小鼠连续9~15d灌喂30mg/kg的雷公藤多甙(TⅡ),以观察该药的抑制效应。结果表明,0.25U的OT2次注射后能明显导致小鼠的DTH;TⅡ应用后可有效地减轻第2次OT注射局部血管的通透性,降低DTH的发生率。通过树突状细胞(DC)过继后对DTH的影响、TⅡ对DC作用的研究及模型小鼠淋巴细胞增殖能力和血清TNF、NO水平的检测,对DTH的发生和TⅡ抑制DTH的机制做了初步探讨。
Comparison of old tuberculin(OT) and bovine serum albumin(BSA) on the effect of inducing delayed type hypersensitivity in mice by detecting the exudation of local blood vessel in OT or BSAinjected area. Using OTinduced DTH model, the inhibiting effect of Tryperygium polyglycoside(TⅡ) was observed by feeding the drug(30 mg/kg) to mice before it was injected with OT. The results showed that 0.25U OT could significantly induce DTH, and DTH response could be prevented by TⅡ. In addition, the mechanisms of DTH response and TⅡ inhibiting effect were discussed by testing the ability of lymphocyte proliferation, the level of TNF and NO in serum, the role of dendritic cell(DC) on DTH and the affect of TⅡ on the function of DC.
