摘要
通过透射电镜观察兔血小板经虎杖晶4号作用后的超微结构变化。为探讨虎杖4号(PD)抑制血小板变形,释放反应机理提供形态学依据。取6只新西兰克,心脏穿刺取血制备富含血小板血浆(PRP),将PRP分为正常对照,生理盐水、吲哚美辛组及3种不同浓度PD实验组。除正常对照组外其余5组均用花生四烯酸(AA)诱导血小板聚集,按超薄切片常规制样,电镜观察各组血小板的超微结构变化,结果证明PD不仅对血小板的变形反应和释放反应有明显的抑制作用而且存在着量效关系。
Objective To provide morphologic basis for the mechanisms by which Polydatin (PD) inhibits platelet aggrega tion. Methods Six New Zealand rabbits were used. Blood was obtained by cardiac puncture. Platelet rich plasma(PRP) was prepared and was divided into 6 groups:control group,normal saline-treated group,indomethacin-treated group,and three dif ferent concentrations of PD-treated groups. Except the control one,all groups were treated with arachidonic acid to induce platelet aggregation. Then the ultrastructural alterations of the platelets were observed. Results and Conclusion PD could prevents platelets from deformation and inhibit the releasing reactions. There is a relationship between dose and efficacy of PD.
