摘要
采用甘油防冻营养液对产精氨酸的黄色短杆菌(Brevibacterium flavum)变异株AN78在普通冰箱冷冻室(-18℃~-20℃)进行冷冻保藏试验,4年中分别进行了AN78菌株的产L-精氨酸试验,在500mL摇瓶试验中产精氨酸30~35g/L;保藏2年的菌种在20L发酵罐试验中产精氨酸63.1g/L,转化率22.8%,发酵周期81h。试验结果好于以前的报道。该方法可作为氨基酸生产行业中一种简便有效的菌种冷冻保藏方法。
The method of freezing preservation with glycerol antifreeze medium for the L-arginine-producing strain of Brevibacterium flavum AN78 in the freezer compartment ( - 18菌 - - 20菌 ) of refrigerator was investigated. During 4 years of freezing preservation, the production d L-arginine by strain AN78 was as high as 30-35g/L in 500 mL shaking flask culture, respectively. The production of L-arginine for freezing preservation of 2 years could reach 63. 1g/L for 81 hours in 20 L fermenter and the conversion rate was 22.8 %. These results were better than that of the report before. Therefore, this method was a simple and effective method of strain preservation for amino acid production.
