甘草酸二铵对大鼠脊髓缺血再灌注损伤后炎症影响的研究 被引量:6

摘要 目的研究甘草酸二铵对大鼠脊髓缺血再灌注损伤后NF-κB、ICAM-1、TNF-α的表达。方法雄性SD大鼠随机分为实验组和对照组,每组40只,夹闭左右肾动脉之间腹主动脉制作脊髓缺血再灌注损伤模型。实验组缺血前10min舌下静脉注射甘草酸二铵20mg/kg,对照组注射同量生理盐水。再灌注3、24、72和168h不同时间点各取10只大鼠腰段脊髓组织,免疫组化法检测NF-κB和ICAM-1表达及ELISA法检测TNF-α的表达。结果3种炎症因子在两组脊髓组织中的表达水平有统计学意义(P<0.05),甘草酸二铵对各时间点脊髓组织中NF-κB表达的抑制率分别是15.40%、20.17%、19.28%、11.11%。结论甘草酸二铵对大鼠脊髓缺血再灌注损伤后NF-κB、ICAM-1和TNF-α表达有抑制作用,抑制了早期炎症反应的发生。 Objective To investigate the effect of Diammonium Glycyrrhizinate for the expression of NF - κB, ICAM - 1 and TNF - α after spinal cord iscbemic - reperfusion injury in rats. Method A model of spinal cord iscbemic - reperfusion injury was completed with intercepting rat's abdominal aorta between right and left renal artery for 30min. 80 male SD rats were randomly divided into experimental group (n = 40) and control group (n = 40), each rat had been administered 20mg/kg DG 10min before isebemia starts via sublingual vein in experimental group and NS in control group. The expression of NF -κB, ICAM - 1 and TNF - α in myeloid tissue was analyzed at different intervals (3, 24, 72 and 168h) respectively after ischemic - reperfusion injury. Result The expression of NF -κB, ICAM - 1 and TNF - α was obviously difference in two groups ( P 〈 0.05 ). The inhabitation ratio of NF - KB by DG at different intervals (3, 24, 72 and 168h) was 15.40 %, 20.17 %, 19.28% and 11.11%, respectively. Conlusion DG has depression effect for the expression of NF - κB, ICAM - 1 and TNF - α, and it has active suppression in earlier period of inflammation occurrence after spinal cord ischemic - reperfusion injury. DG can protect myeloid tissue after ischemia - reperfusion injury.
