摘要
为了研究燕麦麦谷蛋白亚基组成及遗传多样性,本研究采用SDS-PAGE电泳方法对71份不同染色体组型的燕麦材料的麦谷蛋白,进行了电泳分析,并以中国春为参照,按条带迁移距离大小命名,分析条带多态性并以燕麦麦谷蛋白点泳图谱为依据对供试材料进行聚类分析。电泳结果显示燕麦麦谷蛋白主要集中在分子量较低的C区,高分子量麦谷蛋白没有检测到条带。71份燕麦材料共产生20种不同条带,条带g和条带i比较保守,出现的频率分别为64.8%和85.9%。各种条带组合形成62种不同的组合带型,多态性为87.3%。聚类结果显示燕麦麦谷蛋白的电泳谱型与材料的染色体组有很大关系,具有相同染色体组型的同种属的燕麦材料一般先聚为一类,但各材料之间又存在一定的差异。绝大多数六倍体材料在遗传相似性系数约0.68时聚为一类,多数四倍体材料在遗传相似性系数约为0.60时聚为一类,部分二倍体材料在遗传相似性系数约为0.59和0.76时聚为一类。条带a只在六倍体材料AACCDD中出现,而AACC、AABB、CC染色体组的材料中都没有出现,推测可能是D染色体组的特异条带。AsAs和A1A1染色体组存在一定的差异并不完全相同。燕麦麦谷蛋白SDS-PAGE电泳图谱可以作为燕麦指纹图谱。
In order to make good use of oat varieties, the glutenin patterns of 71 oats were analysed by using SDS polyaerylamide gel electrophoresis (SDS-PAGE). Chinese Spring as control, the bands were named according to their electrophoretic mobilities and distribution. The results showed that oats had significant glutenin polymorphism (87.3 % ) ,and among the total 71 materials there existed 20 glutenin bands and 62 different genotypes. The band g and band i appeared higher frequences (64.8 % and 85.9 % ). Compared with Triticum aestivum cv. Chinese Spring, oats possessed the glutenin bands of the lower moleculer weight, mainly existing in the region of B and C. The majority of hexaploid, tetraploid and diploid oats were clustered together at coefficient 0.68 and 0.60, respectively. The partial diploid oats were clustered together at coefficient 0.59 and 0.76. The band a was the special band which was coded by the gene located D. The AsAs and Al Al chromosomes had some differences. It was proved that SDS-PAGE of glutenin could be used asa fingerprint of oats.
