摘要
目的通过检测骨桥蛋白(osteopontin,OPN)在葡萄胎及先兆子痫疾病中的表达情况来探讨OPN在两种疾病中可能参与的发病机制。方法采用免疫组化S-P法及逆转录多聚酶链反应(RT-PCR)检测方法,来比较葡萄胎组织与早孕绒毛、先兆子痫与正常足月胎盘组织中OPN的表达情况。结果各组组织中均有OPNmRNA(245kb)及其蛋白的表达,在相对表达量上,正常早孕绒毛组织中的表达高于葡萄胎组织(P<0.05);正常足月胎盘组织中的表达量也高于先兆子痫患者胎盘组织(P<0.05)。结论OPN在胎盘绒毛组织中的表达差异性,可能与葡萄胎、先兆子痫等滋养层密切相关的妊娠疾病的病理发生过程有一定的关系。
[Objective] To detect the osteopontin (OPN) in the hydatidiform mole or preeclampsia which is closely correlated with trophoblast to explore the role of OPN in pathogenesis of these disease. [Methods] Immunohistochemical staining (S-P method) and reverse transcription polymerase chain reaction (RT-PCR) were used to detect the expression of OPN in tissues of hydatidiform mole, 1st trimester villi, placenta with preeclampsia and normal term placenta. [Remits] OPN were expressed in all tissues. Additionally, the expression levels of OPN were higher in 1st trimester villi than those in hydatidfform mole (P 〈0.05), and the expression levels of OPN in normal term placenta were higher than those in placenta with preeclampsia (P 〈0.05). [Conclusions] The changes of OPN expressions in placenta tissues may be involved in the pathogenesis of trophoblast associated diseases of pregnancy such as hydatidiform mole and preeclampsia.
