摘要
目的:探讨羊富血小板胶-牙周膜成纤维细胞基质(PRgel-periodontal ligament fibroblasts matrix)的合成,并观察将其植入实验动物牙周组织缺损区后促牙周组织再生的可能作用。方法:制备羊牙周组织缺损模型,随机分成PRgel组、PRgel-PDLFs组和空白对照组。采用密度梯度离心法从羊新鲜全血中获取富血小板血浆(platelet-rich plasma,PRP),按一定比例将牛凝血酶和氯化钙加入PRP中合成富血小板胶(platelet-rich gel,PRgel)。体外培养、扩增羊牙周膜成纤维细胞PDLFs,然后将细胞与PRgel培养合成富血小板胶-细胞基质,植入动物牙周组织缺损区。分别于植入后4周和10周处死动物,进行常规组织学检查及CT影像学分析。结果:组织学检查及CT影像学分析显示,3组均在牙根面可见新生牙骨质样组织、牙周膜和牙槽骨样组织的形成,与空白组相比,PRgel-PDLFs组和PRgel组新生牙周组织明显增多,有统计学意义(P<0.01)。PRgel-PDLFs组新生牙周组织量比PRgel组多,有统计学意义(P<0.01)。结论:体外合成的PRgel-PDLFs基质可明显促进牙周组织的再生。
Objective: To study the preparation method of PRgel- periodontal ligament fibroblasts matrix, and to observe its possible function in promoting periodontal tissue regeneration after implanting the matrix into the animal's periodontal defects. Methods: 12 healthy goats were used to establish periodontal defects model. The animals were randomly divided into PRgel group, PRgel-PDLFs group and control group. Goat fresh whole blood obtained from healthy subjects was collected in tubes containing an anticoagulant (heparin). Platelet-rich gel was made by combining the PRP with the thrombin and calcium chloride in certain proportion. The preparation of the matrix was completed through cultured PRgel with periodontal ligament fibroblasts in vitro. The defects were filled with the PRgel-PDLFs or only PRgel. The animals experiment was ended respectively in 4 weeks and 10 weeks after implanting. And specimens were evaluated histomophometrically and analysed by CT image. Results:New cementum, periodontal ligament and alveolar bone can be found on the root surfaces in all three groups. Compared with the blank group, new periodontal tissue in PRgel-PDLFs and PRgel groups have increased obviously, there was obviously statistics significance( P 〈0.01 ). New periodontal tissues in PRgel-PDLFs are more than those in PRgel group, there is statistics significance( P 〈 0.01 ). Conclusion: PRgel-PDLFs matrix can promote periodontal tissue regeneration of periodontal defects. It suggests that this PRgel-PDLFs matrix may play important role in promoting periodontal regeneration.
