摘要
小鼠感染日本血吸虫尾蚴后第7d开始,灌服青蒿琥酯300mg/kg·次,每wk1次,连续4次。在首次给药和第2次给药后的不同时间收集童虫作电镜观察。结果:首次给药后4h,扫描电镜见虫体表皮肿胀;8-12h后表皮褶嵴消失、糜烂;第1d即发现死亡虫体。以后几次给药,病变加重,虫体大部分死亡。透射电镜见皮层细胞肿胀、肌层坏死及溶解、肠腔上皮破坏和脱落。实验表明该药对日本血吸虫童虫有直接损害作用。
NIH mice were infected with cercariae of S. japonicm and divided into two groups (A and B). 7 days after infection,group A were treated with Artesunate one time weekly for 4 times,the dose was 300mg·kg-1. In different time after every adminstration,mice were killed to collect schistosomula, Group B was control. The results showed that 4- 12 hours after first adminstration,obvious tegumental surface alternations were observed by SEM and TEM including swelling, wrinkling and constriction,collapse of sensory papillas. The tegument became thin and the fold dissappeared. One day after adminstration,dead worms were found.After the continuing three administrations,most worms died. Necrosis of large areas of the surface and damage of suckers were found,muscle layers under the tegument dissolved and cells of intestine lumen injured. The results showed that Artesunate may directly damage schistosomula.
