晶状体上皮源生长因子对高浓度葡萄糖诱导凋亡晶状体上皮细胞的保护作用

摘要 目的:研究高浓度葡萄糖诱导人LECs凋亡以及LEDGF对LECs凋亡的影响。方法:采用终浓度为5.56,11.11,22.22mmol/L葡萄糖处理体外培养的LECs,诱导建立LECs凋亡模型。用终浓度为200ng/mL的LEDGF抗体进行干预。利用流式细胞仪对LECs凋亡情况进行检测。结果:流式细胞仪检测发现随着葡萄糖浓度增高,LECs凋亡百分率亦呈升高趋势,不同组间差异有显著性。LEDGF抗体能够增加由高糖引起的LECs凋亡百分率。结论:高浓度葡萄糖可在体外诱导兔LECs凋亡。LEDGF可保护LECs并减少其凋亡的发生。 AIM:To observe whether apoptosis of human lens epithelial cells (LECs) can be induced by high ambient glucose and be affected by LEDGF.METHODS= LECs were exposed to high ambient glucose with the final concentration of 5.56, 11.11, 22.22mmol/L respectively. A model of apoptotic LECs was established and affected by the Ab to LEDGF with the final concentration of 200ng/mL. The apoptosis of LECs was observed and quantified by flow cytometry (FCM~).RESULTS: The percentage of apoptotic LECs was escalated with glucose concentration increasing. There were significant differences between different groups. The percentage of apoptotic LECs increased after using the Ab to LEDGF.CONCLUSION. Apoptosis of LECs can be induced by high ambient glucose. LEDGF protects would LECs and reduce the occurrence of apoptosis.
