3种中药复方对小鼠急性酒精性肝损伤的保护作用

Protective Effect of Three Kinds of Traditional Chinese Herbal Compound on Alcohol-induced Acute Liver Injury in Mice

目的:观察不同组成的中药复方对急性酒精性肝损伤的保护作用,并探讨其可能机制。方法:将小鼠随机分为6组:对照组、模型组、当飞利肝宁组、清肝复方Ⅰ号组、清肝复方Ⅱ号组、清肝复方Ⅲ号组,以35度红星二锅头灌胃10 d复制急性肝损伤小鼠模型,检测小鼠血清谷氨酸转氨酶(glutamate transaminase,ALT)、天冬氨酸转氨酶(aspartate aminotransferase,AST)、三酰甘油(triacylglycerol,TG)、γ-谷氨酰转移酶(γ-glutamyl transpeptidase,γ-GT)活性,肝组织丙二醛(ma-londialdehyde,MDA)、谷胱甘肽(glutathione,GSH)、乙醇脱氢酶(ethanol dehydrogenase,ADH)含量,采用光学显微镜观察肝组织形态学变化。结果:中药治疗组能显著抑制小鼠血清ALT、AST、TG以及γ-GT活性的升高(P<0.05),抑制肝组织MDA含量升高(P<0.05),提高肝组织中GSH活性(P<0.05),减轻肝脏的病理损伤。结论:3组中药复方对乙醇引起的小鼠急性肝损伤都具有一定的保护作用,并且以清肝复方Ⅲ的肝脏保护作用最好。

Objective： To observe the protective effects of traditional Chinese prescription on hepatic injury induced by alcohol in mice, and explores its mechanism. Methods： The Kunming mice were divided into six groups： control group, model group, Qinggan Compound Ⅰgroup, Qinggan Compound Ⅱ group, Qinggan Compound Ⅲ group and Dangfei liganning Capsule group. The liver injury model was in duced by consecutive intragastric injection of 35 degrees Hongxing Er Guotou wine during 10 days. The activities of serum alanine aminotransferase （ALT）, aspartate aminotransferase （AST）, triglyceride （TG） and γ-glutamyl transpeptidase （γ-GT）,the contents of malondialdehyde （MDA）and the activities of gluta- thione （GSH） in hepatic tissues were measured. The histological changes of liver were observed by optical microscope. Results： The three kinds of traditional Chinese herbal compound significantly inhibited the activities of ALT. AST. TG and γ-GT in serum（P〈0.05）, and decreased the content of MDA（P〈0.05）, meanwhile increased the activities of GSH in hepatic tissues. Furthermore, the pathological injury was also significantly alleviated in the Qinggan Compound Ⅲ group. Conclusion： The three kinds of traditional Chi nese herbal compound show protective effect on the acute hepatic injury induced by alcohol. Qinggan Corn pound Ⅲ has the best effect on acute alcoholic liver injury.
