摘要
目的:观察氧化应激对非酒精性脂肪性肝炎(NASH)发病的影响,评价枳椇子水提取液对NASH的预防作用。方法:用高脂饲料诱导NASH大鼠动物模型,同时给予枳椇子水提取液进行预防。观察枳椇子水提液对NASH模型大鼠肝组织丙二醛(MDA)、超氧化物歧化酶(SOD)及谷胱甘肽氧化酶(GSH-PX)的影响及肝组织的病理学变化。结果:12周后,模型组肝匀浆中MDA含量明显高于正常组,SOD、GSH-PX呈下降趋势;肝组织呈脂肪变性和坏死性炎症改变。枳椇子水提取液可以明显减轻模型大鼠肝脏炎症和脂肪变程度(P<0.01),降低肝组织MDA(P<0.01),提高肝组织SOD、GSH-PX活性(P<0.01或P<0.05)。结论:氧化应激在非酒精性脂肪性肝炎的发生和发展中起重要作用;枳椇子水提取液能够减轻NASH模型大鼠肝组织的脂质沉积和炎性浸润,对NASH大鼠具有良好的预防作用。
Objective :To observe the effect of oxidation strass on the onset of nonalcoholic stratoheatitis (NASH) and to investigate the effect of hovena dulcis thumb water extract on the prevention of NASH. Methods: Experimental NASH model was established in sixty SD rats with a high fat diet. Hovena dulcis thumb water extract was simultaneously administered. Animal liver tissue samples were collected to observe the changes in Malondialdehyd (MDA), superoxide dismutase, and glutathione peroxidese (GSH-Px) and histology. Results: Compared to the control group, the NASH group had higher MDA and lower SOD and GSH - Px; steatosis and necro - inflammation were observed histologically. The hovena dulcis thumb water extract greatly attenuated steatosis and inflammation(P 〈 0.01 ), decreased MDA( P 〈 0.01 ) , and increased SOD and GSH -Px (P 〈 0.01 ,P 〈 0.05 )in the liver tissues compared to the NASH group. Conclusion: Oxidation stress plays an important role in development of NASH. The hovena dulcis thumb water extract may prevent the occurrence of NASH by improving the pathological changes in the liver tissue.
