摘要
以V型胶原酶分离胰腺得到胰岛,在EGTA预孵育后以低浓度的胰蛋白酶和DNaseI酶联用消化胰岛,建立了大鼠胰腺β细胞的分离及原代培养方法.反复实验表明(n=10),所获得的细胞表面光洁、完整、呈球形,功能正常,能对生理剌激物迅速反应,适用于通常的单个细胞的电生理学实验.
A practical method including following steps for the isolation and primary culture of rat pancreatic β cells has been developed: isolating islets from rat pancreas with collagenase type V; incubating islets with EGTA; digesting islets with low concentration of trypsin and DNase I; and then centrifugalizing, washing, collecting and culturing single cells. Repeated experiments ( n =10) show that the cells obtained are clear, intact and spherical. Their normal function is verified by their quick responses to physiological stimuli. The method is suitable for usual electrophysiological experiment of single cell.
