摘要
目的探讨绿茶提取物EGCG对人肝癌细胞株SMMC-7721体外增殖和凋亡的影响,预测其对人肝癌的抗癌作用。方法体外培养人肝癌SMMC-7721细胞,采用不同浓度的EGCG对其进行干预,用MTT法观测其对肝癌细胞增殖能力的影响;用免疫细胞化学法检测肝癌细胞中Ki67的表达变化;用流式细胞仪检测其对肝癌细胞凋亡的影响。结果在EGCG的作用下,SMMC-7721细胞增殖能力明显受到抑制(P<0.05),Ki67表达水平下调(P<0.05),细胞凋亡明显增加(P<0.05)。结论EGCG可能通过增加细胞凋亡,抑制Ki67表达达到抗癌作用。
Objective To confirm the effect of EC, CG on the proliferation ability and apoptosis of hepatoma carcinoma cell llne SMMG7721.Methods The proliferation ability of cultured SMMC-7721 cells in varying concentration of EC, CG were tested by MTT analysis, the expression Of Ki67 were tested with immunocytocheraical method and cell apoptosis were detected with flow cytometry. Results In EGCG,the proliferation ability of SMMC-7721 cells was inhibited( P 〈 0.05) and the expression of Ki67 was downregulated (P 〈 0.05), the apoptosis was raised( P 〈 0.051. Conclusion EGCG inhibits the proliferation of SMMC-7721 cells, downregulates the expression of Ki67 as well as increases the apoptosis, which achieves its anti-hepatoma carcinoma effect.
