摘要
对Arthrobacter chlorophenolicus GS0202菌产人参皂苷糖苷酶进行分离纯化并对其性质进行了研究。结果表明,电泳纯人参皂苷糖苷酶的分子质量约为66ku,最适反应pH值为5.0,最适温度为40℃,在pH3.0~7.0,20~60℃酶活力相对稳定。K+、Na+、Zn2+对酶活性影响很小;Ca2+、Mg2+、Cu2+对酶活性有明显的抑制作用。Km值为4.392mmol/L,Vmax值为0.1275mmol/(h.L)。
The paper was aimed to investigate panaxsaponin-glucosidase which was purified from the broth of microbion (Arthrobacter chlorophenolicus GS0202 ) and its properties. The results showed that the molecular weight of which was 66ku,the optimum pH and temperature were pH 5.0 and 40℃ respectively,the enzyme was stable in the pH range from 3.0 to 7.0 and the temperature ranges from 20℃to 60℃. K^+ , Na^+ and Zn^2+ had little contribution to the enzyme; Ca^2+ , Mg^2+ and Cu^2+ showed noticeable inhibiting attributes. The kinetic analysis revealed that the value of Km and VmAx was 4. 392mmol/L and 0. 1275mmol/(h · L) , respectively.
