摘要
金针菇20642菌株,深层培养所得的菌丝体经热水浸取、盐析、透析、柱层析分离纯化得到一种糖蛋白(FMGP),含蛋白质796%,糖67%,糖组成分为半乳糖、葡萄糖、甘露糖和一种未知的糖;蛋白质由18种氨基酸组成,以天冬氨酸和谷氨酸的含量最高;红外光谱显示,在1530cm-1,1620cm-1,3420cm-1处有三个明显的吸收高峰。经HPLC示差检测器测定,其平均分子量为348Kd。
A glycoprotein has been isolated through hot water extract,salting out,dialysis,columr chromatography from the submerged culture of mycelia of Flammulina velutipes “20642”.Its protein content is 79 6% and its carbohydrate cotent is 6 7%.The protein is made of 18 types of amino acids.The content of aspartic acids and glutamic acids are the highest.The carbohydrates are composed as galactose,glucose,mannose,and an unknown sugar.Infrared absorption spectrum reveals that it has the absorption peaks at 1 530 cm -1 , 1 620 cm -1 , 83 420 cm -1 . With HPLC analysis and with the know molecular weight of dextran as the standard,the average molecular weight 348Kd.This glycoprotein is like Japanese proflamin.
