多药耐药铜绿假单胞菌耐药基因研究 被引量:3

摘要 目的研究1株多药耐药铜绿假单胞菌的β-内酰胺酶基因、菌膜蛋白OprD_2基因、整合子和转座子介导的各种耐药基因、16S rRNA甲基化酶及氨基糖苷类修饰基因以及氯霉素与四环素相关耐药基因的的存在情况。方法以K-B法测量其对抗菌药物的耐药性;以PCR法检测其相关耐药基因;以PCR直接全自动荧光法进行阳性基因测序。结果该株铜绿假单胞菌检出TEM基因OXA-10基因,且伴有OprD_2缺失;转座子遗传标记MerA阳性,Ⅰ类整合子遗传标记qacE△1-Sul1阳性;耐氨基糖苷类基因aac(3)-Ⅱ、aac(6′)-Ⅱ、rmtB阳性,耐氯霉素基因cmlA阳性。结论造成本株铜绿假单胞菌多药耐药的主要原因是该菌株存在多种抗生素相关耐药基因。 Objective To explore the situation of β-lactamase gene, oprD2 gene of outer mem- brane protein, various drug resistance genes mediated by integron and transposon, 16S rRNA methy- lase gene and aminoglycoside modifying enzyme gene, as well as chloramphenicol- and tetracycline-re- lated resistance genes in 1 strain of multidrug resistant Pseudomonas aeruginosa. Methods The antibi- otics susceptibility was tested by K-B method; the drug resistance related genes were detected by poly- merase chain reaction (PCR) ; the positive genes were ampilified and sequenced by PCR fluorescence spectrophotometry. Results The genes of TEM type OXA-10, qacEA1-Sull, aac(3)-Ⅱ , aac(6')-Ⅱ , rmtB and cmlA were found in the Pseudomonas aeruginosa strain, and OprD2 was negative. Conclusion The main reason for the Pseudomonas aeruginosa strain drug resistance is existence of muhi-antiobiotic related drug resistance genes in the Pseudomonas aeruginosa strain.
