摘要
目的:为探讨曲尼司特预防再狭窄的可能性,研究了曲尼司特对培养的大鼠主动脉平滑肌细胞(ASMC)增殖的影响。方法:以培养的大鼠ASMC为模型,应用^3H-TdR掺入试验观察ASMC增殖情况。结果:曲尼司特呈剂量信赖性无抑制基础、血小板源生长因子(PDGF)和血管紧张素Ⅱ(AngⅡ)诱导的ASM DNA合成,与相应对照组比较P〈0.05或P〈0.01。结论:曲尼司特能明显抑制ASMC增殖。
Objective:To investigate the possibility of tranilast on the prevention of restenosis,we examined the ef- fect of tranilast on the proliferation of rat aortic smooth muscle cell(ASMC) cultured in vitro.Methods:Rat ASMC mod- el cultured in vitro was established.H-thymidine(~3H-TdR)incorporation test was employed to detect proliferation of ASMC.Results:Tranilast inhibited basal rat ASMC DNA synthesis,also markedly blunted both platelet-derived growth factor(PDGF) and Angiotensin Ⅱ(Ang Ⅱ)which induced increase of ASMC DNA synthesis in a dose-dependent manner (P<0.05 or 0.01 vs control).Conclusion:Tranilast can markedly inhibit ASMC proliferation,and may be beneficial in the prevention of restenosis.
