摘要
应用荧光原位杂交技术观察顺铂诱导膀胱癌细胞株BIU87体外细胞凋亡过程中野生型P53和CHaras基因的表达,探讨抗癌药物诱导膀胱癌细胞凋亡过程中抗癌基因和癌基因的作用。结果显示:顺铂能够明显地诱导膀胱癌细胞凋亡,具有一定范围内的作用时间和药物浓度的依赖性。随凋亡细胞数上升,野生型P53表达增强,而CHaras基因表达下降,提示膀胱癌细胞凋亡可能是顺铂的杀瘤效应途径之一。P53和CHaras基因均参与了细胞凋亡的调控过程,野生型P53基因可能是通过下调CHaras基因的表达而发挥其抑制细胞增殖活性,促进凋亡的发生。
The expression of P53 and CHaras of BIU87 bladder cancer cell lines with apoptosis induced by CDDP was detected by means of fluorescence immuno in situ hybridization (FISH).An extensive apoptosis in BIU87 cells treated by CDDP has occured, which was dependent on an apropriate concentration of the drug or treatment time. The positive expression rate of P53 has been significantly upregulated and CHaras was downregulated as apoptotic index increased. These results suggested enhanced expression of P53 or decreased expression of CHaras might be involved in apoptosis or inhibition of the proliferation of bladder cancer cells induced by CDDP.
