细胞因子对甲状腺相关性免疫眼眶病发病机制的研究 被引量:5

A study of cytokines in pathogenesis of thyroid-related immune orbitopathy

摘要 目的探讨细胞因子在甲状腺相关性免疫眼眶病(thyroidrelatedimmuneorbitopathy,TRIO)发病机制中的作用。方法体外培养眶成纤维细胞,应用免疫组化,观察细胞因子对眶成纤维细胞人类白细胞表面抗原Ⅱ(humanleukocyteantigenⅡ,HLAⅡ)异常表达的诱导作用。结果干扰素γ(interferonγ,IFNγ)、白细胞介素1(interleukin1,IL1)及肿瘤坏死因子(tumornecrosisfactor,TNF)对眶成纤维细胞HLAⅡ抗原异常表达有诱导作用,IFNγ的诱导作用最强且与时间和浓度呈正相关。结论细胞因子(IFNγ,IL1,TNF等)可诱导眶成纤维细胞HLAⅡ抗原异常表达,在启动免疫反应及甲状腺相关性免疫眼眶病发病机制中起重要作用。 Objective To explore the pathogenesis of thyroidrelated immune orbitopathy (TRIO). Methods Human orbital fibroblasts were cultured in vitro and immunohistochemical techniques were used to study cytokines that induce effect on HLAII antigen expression of orbital fibroblasts. Results It is shown that interferonγ (IFNγ), interleukin1 (IL1) and tumor necrosis factor (TNF) could induce the expression of orbital fibroblast HLAII antigens. The induced effect of IFNγ is of good linear correlations with time and concentration. Conclusion HLAII antigen expression can be induced by IFNγ, IL1 and TNF. In the course of immune reaction and pathogenesis of TRIO, cytokines play important roles. 
