毒素清对细菌性肺炎痰热证肺组织炎性因子的影响 被引量:6

Effect of Dusuqing on the Inflammatory Factors in the Lung Tissue of Bacterial Pneumonia and Heat-phelgm Combining with Disease and Syndrome

摘要 目的:从支气管肺泡灌洗液(BALF)中肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)、细胞间黏附分子-1(ICAM-1)的变化揭示中药毒素清对细菌性肺炎痰热证大鼠肺组织的保护作用机制。方法:取W istar大鼠随机分为假手术组、肺炎组、模型组、对照治疗组、毒素清组。制备细菌性肺炎痰热证模型。观察肺组织损伤、外周血白细胞(WBC)、中性粒细胞(PMN)计数及BALF中TNFα-、IL-6、ICAM-1的水平。结果:与假手术组相比,模型组肺组织损伤最明显;模型组外周血WBC、PMN和BALF中TNF-α、IL-6、ICAM-1水平较假手术组明显增加(P<0.01);与模型组相比,毒素清组、对照治疗组肺组织病理损伤减轻,毒素清尤为明显,毒素清组、对照治疗组外周血WBC、PMN及BALF中TNF-α、IL-6、ICAM-1水平均降低,毒素清组降低更明显(P<0.01)。结论:炎性因子TNF-α、IL-6、ICAM-1参与细菌性肺炎痰热证肺组织损伤的发生发展,抑制TNF-α、IL-6、ICAM-1的表达可能是毒素清保护细菌性肺炎痰热证肺组织损伤的主要途径。 Objective: To study the protective effect of Dusuqing(DSQ) of Chinese medicine on the lung tissue of rats from the level changes of tumor necrosis factor-α( TNF -α), interleukiu-6 ( IL - 6 ) and intercellular cell adhesion molecule - 1 ( ICAM - 1 ). Methods:The wistar rats was randomly divided into five groups, the sham-operated group, the bacterial pneumia group, the model group,the control-treated group and the DSQ group. The animal model of bacterial pneumonia was established, and on the basis of which, wind and heat was continuously administered. The pathological injury of lung tissue, white blood cells (WBC), neutrophilic granulocyte, and the levels of TNF -α, IL-6 and ICAM-1 in bronchoalveolar lavage fluid(BALF) was measured. Results: Compared with the sham-operated group,the patho-damage in model group was more severe. The counts of WBC and PMN and levels of TNF-α,IL-6 and ICAM-1 in BALF in model group were evidently more than in sham-operated group. The degree of lung tissue injury, the contents of WBC and PMN of peripheral blood, and the levels of TNF-α, IL-6 and ICAM-1 in BALF in the model group was more higher than those of the sham-operation group( P 〈 0. 01 ). The pathological injury of lung tissue in the DSQ group and the treated group was mere lesser than in the model group,the lesser in the DSQ group was more evident than in the control- treated group. The pathologic change in the DSQ group and the treated group was mere lesser than in the model group,the lesser in the DSQ group was more evident than in the control - treated group. The contents of WBC and PMN of peripheral blood, and the levels of TNF-α, IL-6 and ICAM-1 in BALF in the DSQ group and the treated group was mere lesser than in the model group, the decrease in the DSQ group was more obvisous than in the control-treated group( P 〈0. 01 ). Conclusion :DSQ showed an obvious effect in protecting the lung tissue of bacterial pneumonia and heat-phelgm combining with disease and syndrome. The mechanism might be related with the lowering of inflammatory cytokines such as oversecreted TNF-α,IL-6 and ICAM-1.
