摘要
比较了不同有机溶剂对锦鲤鳞色素的提取效果和色素在不同溶剂中的稳定性。试验分别以乙醚、丙酮、乙醇和石油醚作为提取溶剂,采用振荡浸提法提取(30℃下提取3 h)锦鲤的鱼鳞色素,并于340 nm下测定了色素提取液的吸光度。结果表明,提取相同质量的鱼鳞色素,乙醚和乙醇提取液的吸光度较大,石油醚提取液的吸光度最小。在日光密闭保存24 h和48 h后,乙醇中色素的破坏率均显著低于乙醚、丙酮和石油醚中,表明乙醇更适宜作为提取鱼鳞色素的溶剂。
Comparison of extracting efficiencies of scale pigments and stability of pigments in different organic solvents were carried out in colored common carp (Cyprinus carpio) by different organic solvents. The scale pigments were extracted by shaking with different solvents (ether, acetone, ethanol and petroleum ether) for 3 h at 30 ℃, and the absorbance of extracting solution was determined at wavelength of 340 nm. For the scales with the same weight, there was higher absorbance in ether and ethanol solution than that in acetone and petroleum solutions. The maximal absorbance value was found in the petroleum ether solution. There was significantly lower damage rate of pigments in ethanol than that in aether, acetone and petroleum ether for 24 h or 48 h under the condition of illumination and obturation, indicating that ethanol was more appropriate for the extracting solvent for total pigments in fish scales.
