摘要
目的研究系统性红斑狼疮(SLE)患者外周血T细胞中磷酸化ERM(ezrin—radixin—moesin)蛋白表达情况并探讨其临床意义.方法收集本院门诊或住院的SLE患者27例和类风湿关节炎(RA)患者5例,正常健康人12例作为对照组。采用免疫印迹法和免疫荧光法检测外周血T细胞磷酸化ERM蛋白,并比较各组间磷酸化ERM蛋白表达的差异。比较SLE患者T细胞体外培养24h后、SLE患者新鲜分离的T细胞及对照组T细胞的磷酸化ERM蛋白表达的差异。分析SLE患者T细胞磷酸化ERM蛋白表达与SLE患者SLE疾病活动指数(SLEDAI)的关系。结果新鲜分离的SLE患者外周血T细胞磷酸化ERM蛋白相对表达量高于对照组和RA组(均P〈0.05)。与对照组比较,SLE患者T细胞在体外培养24h后磷酸化ERM蛋白相对表达量差异无统计学意义(P〉0.05)。中、重度病情活动的SLE患者外周血T细胞磷酸化ERM蛋白相对表达量显著高于轻度病情活动者(0.61±0.16比0.44±0.07,0.63±0.15比0.44±0.07,均P〈0.05)。结论SLE患者T细胞存在磷酸化ERM蛋白表达异常,并且可能与狼疮疾病活动度有关。
Objective To detect the expression of phosphorylated ERM (ezrin/radixin/moesin) protein in peripheral blood T cells from patients with systemic lupus erythematosus (SLE), and to elucidate its clinical significance. Methods Twenty-seven patients with SLE and 5 patieuts with rheumatoid arthritis (RA) from outpatient or inpatient department in our hospital were recruited. And 12 healthy people were enrolled as control group. Phosphorylated ERM protein was assessed by Western blot analysis and immunofluorescent microscopy respectively, and the expression differences were compared among 3 groups. Phosphorylated ERM protein expressions of 24 h after T cell cultures in vitro, freshly isolated T cell from SLE patients and T cells from control group were compared. The relationship between phosphorylated ERM protein expression and SLE disease activity index (SLEDAI) was analyzed. Results Compared with control group and RA group, the expression of phosphorylated ERM protein in freshly isolated peripheral blood T cell from SLE patients was significantly increased (all P〈0.05). The phosphorylated ERM protein expression of T cell cultured in vitro for 24 h was not different with that of the control group (P〉0.05). Compared with the slight disease activity patients with SLE, phosphorylated ERM protein expressions of peripheral blood T cell in the moderate or severe activity patients with SLE were significantly increased (0.61 ±0.16 vs 0.44±0.07, 0.63± 0.15 vs 0.44±0.07, all P〈0.05 ). Conclusion Phosphorylated ERM protein of T cells expresses abnornaly in patients with SLE which may be associated with the disease activity of SLE.
