摘要
目的:通过研究清肝活血方及其拆方对酒精性肝病大鼠肝组织P-ERK、NF-κB蛋白表达的影响,探讨清肝活血方及其拆方治疗酒精性肝病的作用机制。方法:将Wistar大鼠随机分为空白组、CCl4组、造模组,造模组采用复合因素复制酒精性肝病模型。4周后将造模组随机分为模型组、清肝方组、活血方组、清肝活血方组。各中药组予相应中药灌胃2周。观察肝功能、肝脏病理学变化、Western blot法检测肝组织P-ERK、NF-κB蛋白表达。结果:清肝活血方及其拆方均可明显改善模型大鼠肝脏脂肪变性及炎症程度,清肝活血方优于拆方。清肝活血方能显著降低模型大鼠血清ALT水平,拆方无明显作用;清肝活血方及其拆方均可显著降低模型大鼠血清AST水平及肝组织P-ERK、NF-κB蛋白表达。结论:清肝活血方及其拆方可通过抑制肝组织P-ERK、NF-κB表达发挥对酒精性肝病的防治作用,清肝活血方的作用优于拆方。
Objective : To study the action mechanism of Qingganhuoxue Recipe (QGHXR) and its separated recipes by research its effect on the expression of P - ERK and NF - κB in rats with alcohol liver disease(ALD). Methods : Wistar rats were divided into three groups randomly:control group, CCl4 group and experiment group, experiment group was disposaled with composite factor Four weeks later, the experiment group was divided into 4 groups randomly: QGHXR group ,Qinggan recipe group(QGR) ,Huoxue recipe group(HXR) and model group. All rats were given suitable drugs two weeks. All rats were sacrificed at the end of the sixth week. The levels of ALT, AST in the serum were analyzed. Pathological changes in liver tissues were observed. The expression of P - ERK and NF - κB were evaluated with Western blot. Results : QGHXR and its separated recipes improved the degree of liver steatosis and inflammation. The effect of QGHXR is best in them. QGHXR degraded the level of ALT in the blood of model rats,but its separated recipes did not show manifest effect. QGHXR and its separated recipes degraded the level of AST and the expression of P - ERK significantly. Conclusion: QGHXR and its separated recipes may provide protection against ALD in rats through decreasing the expression of P - ERK and NF - κB. The effect of QGHXR is best in them.
