大鼠肠缺血-再灌注早期多个器官内细胞凋亡的研究 被引量:10

Study of Apoptosis in Multiple Organs in Rats at Early Stage of Intestinal Ischemia Reperfusion

摘要 目的探讨细胞凋亡在多器官功能障碍综合征发病中的作用。方法夹闭和放松大鼠肠系膜上动脉复制肠缺血-再灌注模型;显微镜检测分别用HE、吖啶橙-溴化乙锭、Gomori银-HE染色的肠、肝、肾组织的连续切片,观察3个器官组织内细胞凋亡的分布及变化;分离缺血-再灌注不同时间组的肝细胞、流式细胞仪检测肝细胞凋亡百分率。结果(1)大鼠肠缺血-再灌注早期(术后3小时内),肝、肠、肾内凋亡的实质细胞都呈多部位局灶性分布的特点。(2)肝细胞凋亡百分率随缺血-再灌注时间延长而升高(P<0.001),且显著高于假手术组(P<0.001)。结论肠缺血-再灌注早期多个器官组织内细胞凋亡相当广泛。 Aim To elucidate the probable roles of apoptosis in the pathogenesis of multiple organ dysfunction syndrome. Methods A rat ischemia reperfusion model reproduced by clipping and releasing the superior mesenteric artery. Microscoping sequential tissue slices of small intestine, liver and kidney were stained with HE, acridine orange and ethidium bromide, or Gormori's silver HE, respectively. And the percentage of apoptosis in hepatocytes isolated from different ischemia reperfusion time points using flow cytometry was detected. Results (1) In the early phase of intestinal ischemia reperfusion (3 hours after operation), apoptotic parenchymal cells in the small intestine, liver and kidney were distributed multifocally. (2) Apoptotic percentages of hepatocytes increased with the time of ischemia reperfusion ( P <0.001), and statistically higher than that of the sham operation groups. Conclusion At the early stage of intestinal ischemia reperfusion, apoptotic cells in the tissues of multi organs are very common. And this kind of cell death may play an important role in the pathogenesis of multiple organ dysfunction syndrome.
