摘要
试验利用通用引物ITS1和ITS4对南瓜疫病菌(Phytophthora capsici)的19个菌株的ITS rDNA进行扩增,然后利用4种限制性内切酶酶切PCR扩增产物,分析各菌株间的DNA限制性片段多态性。运用ITS通用引物在所有供试南瓜疫病菌菌株上均可扩增到一条长为575bp的特异DNA片段。经限制性内切酶酶切和聚类分析,将黑龙江省南瓜疫病菌可划分为4种病原型。运用一对ITS通用引物扩增供试菌株得到20个ITS标记,其中多态性标记占95%。供试菌株在遗传上有相似性,差异也非常明显,表明南瓜疫病菌群体内部存在着丰富的遗传多样性。
In this paper, by universal primers ITS1 and ITS4 the pumpkin Phytophthora capsici of the 19 strains of the ITS rDNA were amplified, and then used 4 restriction enzymes (Hinf I, Hae Ⅲ, EcoR I, Hind Ⅲ) to cut PCR amplification products, analysis of the strains between the DNA restriction fragment polymorphism. ITS using of universal primers for all bacteria strains of the disease tested on the pumpkin could be amplified to a length of 575 bp specific DNA fragment. By the restriction enzyme cuting and cluster analysis, Heilongjiang Province pumpkin Phytophthora pathogen could be divided into 4 types. The use of a pair of common ITS amplified strains tested had 20 ITS tags, which accounted for 95% of the polymorphic markers. Isolates had similarities in the genetic there, the difference was very obvious, it concluded that pumpkin strain of the disease within groups was abundant genetic diversity.
