百草枯对大鼠肾损伤作用

摘要 目的探讨百草枯(PQ)对肾损伤中诱导型一氧化氮合酶(iNOS)和血红素氧合酶-1(HO-1)作用机制。方法84只SD大鼠随机分为对照组和染毒组,每组42只。染毒组腹腔注射百草枯25 mg/kg,对照组给予等量生理盐水。分别于给药后3,6,12 h,1,2,3,5 d观察肾组织病理学变化及HO-1、HO-1 mRNA、iNOS表达变化。结果与对照组比较,染毒组于染毒3 h肾组织见充血、水肿及空泡变性等变化;染毒3 h后iNOS与HO-1呈阳性表达,染毒1 d后达高峰,平均分分别为(8±2.23)和(7±2),明显高于对照组(P<0.05),染毒3 h后HO-1mRNA表达水平为(0.53±0.21),明显高于对照组的(0.3±0.12)(P<0.05),之后表达逐渐减弱,至染毒后5 d差异无统计学意义。结论iNOS与HO-1与PQ染毒肾损伤过程有关。 Objective To investigate the relation of paraquart-induced renal injury and the changes of heme oxygenase-1(HO-1) expression and inducible nitric oxide synthase(iNOS).Methods Eighty-four adult healthy Spragne-Dawley rats were randomly divided into two groups,the control group(n=42) and the paraquart(PQ) poisoning group(n=42).The rats in the paraquart poisoning group were treated with 25 mg/kg PQ intraperitoneally and the rats in the control group were treated with the same dose of normal saline.Then the histopathological change,the expression of iNOS,HO-1mRNA were observed.Results Compared with the control group,there were obvious lesions in renal tubule of cortical part,including congestion,edema and vacuolar degeneration in the paraquart poisioning group.These pathologic changes gradually reached the peak on the 1st day,and did not relieved till the end of the study.Compared with the control group,the expression of iNOS in the paraquart poisoning group was observed at 3rd hour,reached the pesk on the 1st day with an immunohistochemistry score of 8±2.23,and continued at a high level.Compared with the control group,the expression of HO-1 in the paraquart poisoning group was observed at the 3rd hour with an immunohisochemistry score(IHS) of 3.33±1.75(P〈0.05).The expression of HO-1mRNA increased and reached the peak on the 1st day with a INS score of 7±2(P〈0.05).Conclusion HO-1 and iNOS were involved in the procees of renal injury induced by paraquart.
