鼠β-防御素2抗宫颈癌的实验研究 被引量:2

摘要 目的:建立BALB/c小鼠U14宫颈癌移植瘤模型,观察肿瘤生长情况,探讨mBD2对小鼠机体免疫功能的影响。方法:采用无内毒素质粒大抽试剂盒抽提pcDNA3.1(+)/mBD2、pcDNA3.1(+)/rmBD2和pcDNA3.1(+)质粒DNA。将荷瘤小鼠随机分为5组,分别采用pcDNA3.1(+)/mBD2、pcDNA3.1(+)/rmBD2质粒治疗,同时设立pcDNA3.1(+)、顺铂和PBS组对照;观察肿瘤生长状况、荷瘤小鼠存活时间,绘制小鼠生存曲线。结果:成功建立了BALB/c小鼠U14移植瘤模型。采用pcDNA3.1(+)/mBD2、pcDNA3.1(+)/rmBD2、pcDNA3.1(+)治疗造模成功的荷瘤小鼠,结果显示:pcDNA组及PBS组肿瘤生长迅速,出现大范围出血、坏死,并于治疗后7d全部死亡;mBD2组与rmBD2组小鼠肿瘤生长相对缓慢,出现局部出血坏死等现象,至治疗2周,出现死亡,至治疗结束时,每组仍有3只小鼠存活,而Pt组小鼠肿瘤生长慢,小鼠生长较为活跃,至治疗21d,开始出现死亡,pcDNA和PBS组与mBD2组和rmBD2组及Pt组与mBD2组和rmBD2组生存时间比较,结果有统计学意义(P<0.05),mBD2组和rmBD2生存时间比较,也具有统计学意义(P<0.05)。结论:mBD2能够抑制移植瘤的生长,延长荷瘤小鼠的生存时间。在抑瘤效果上,mBD2优于外加信号肽重组mBD2作用。
